Principle of Targeting via Small Molecules
The principle of targeting via small molecules is to target the molecular biology basis of tumorigenesis, usually to regulate the activity of protein targets. The protein targets mainly include enzymes and receptors. These small molecules can be divided into receptor agonists and receptor antagonists while acting on the receptor. Small molecules that act as inhibitors of enzymes reduce their catalytic activity by binding to them.
Some Examples of Bio-Targeted Small Molecules
These small molecules can bind with the highly expressed targets on the surface of cancer cells for active targeting in drug delivery.
1. Folic acid (folate). Cancer and embryonic cells preferentially use the folic acid receptors (FR) that have high affinity for folic acid to absorb folate via receptor-mediated endocytosis, which helps folate become an attractive targeted ligand for active targeting.
2. Glucose transporter targeting agent glufosfamide. Cancer cells highly rely on glucose and glycolysis for survival and express glucose transporters.
